Accuris qMax cDNA Synthesis Kit and qMax 1st Strand cDNA Synthesis Flex Kit allow the user to generate high yields of cDNA from low amounts of RNA starting template (total RNA or purified mRNA), via reverse transcription. cDNA is then used for further experimentation and analysis (e.g. Quantitative PCR).

The qMax cDNA Synthesis Kit contains two components:

An optimized 5X buffer that contains anchored oligo (dT) primers, random hexamers, dNTPs, 20mM MgCl, stabilizers and enhancers

20X Reverse Transcriptase (an RNA-dependant DNA polymerase, derived from MMLV, and an RNase Inhibitor)

→Works optimally with 4 pg to 0.5 ug of total RNA or oligo(dT) purified mRNA

The qMax 1st Strand cDNA Synthesis Flex Kit contains four components:

An optimized 5X buffer that contains dNTPs, 20mM MgCl, stabilizers and enhancers

20X Reverse Transcriptase (an RNA-dependant DNA polymerase, derived from MMLV, and an RNase Inhibitor)

Solution of oligo (dT) primers

Solution of random hexamer primers

→Works optimally with 10 pg to 2.0 ug of total RNA or 5 pg to 0.5 ug of oligo (dT) purified mRNA

→qMax Reverse Transcriptase is quite thermostable and will allow for reverse transcription at a higher temperature (45°C), which helps alleviate problems caused by RNA secondary structure formation.

→Anchored oligo (dT) primers will bind to the very 3′ end of the mRNA, preventing the amplification of truncated cDNAs by not binding to internal poly(A) sites.

→Separate solutions of oligo (dT) primers and random hexamer primers gives the user more flexibility in protocol design.

Amplified DNA is detected by means of fluorogenic-labeled probes such as TaqMan®, Scorpions® and molecular beacons, all of which the qMax Probe qPCR Mix is compatible for use with. Works efficiently with singleplex or multiplex reaction setups.

Uses genomic DNA (gDNA) or complimentary DNA (cDNA) as starting template for amplification of gene target(s) of interest.

Using a PCR master mix makes reaction setup simpler and reduces the chances of contamination (e.g. less pipetting from more tubes). Also it keeps future reaction compositions uniform to past reaction compositions (i.e. reactions set up the next day, for example, will be uniform to reactions setup the previous day because the reaction mix pipetted, the master mix, is the same).

qPCR, also known as Real-Time PCR, allows the user to amplify DNA by means of PCR and monitor the progression of amplification quantitatively, in real time. That is, qPCR allows the user to measure quantities of amplified target DNA, cycle by cycle.

qMax Probe qPCR Mix comes as a 2X Master Mix that contains Accuris Hot Start Taq Polymerase, dNTPs and buffer that includes PCR enhancers.

Forward and reverse primers, fluorogenic-labeled probes, template DNA and PCR grade water are to be acquired and added to the reaction mix by the user.

→Available with no, low or high levels of ROX (select in dropdown menu). ROX is a fluorescent, passive reference dye that is used to normalize the signal emitted from the reporter dye, linked to the probe(s) of choice, during qPCR.

The fluorescent signal from the passive dye remains constant during qPCR, while the signal from the reporter dye changes as cycles progress. Real-Time PCR instruments have varying requirements for ROX levels.

Two-step reaction (i.e. Reverse Transcription plus DNA Amplification) is performed in one reaction tube with a single reaction setup.

Amplified DNA is detected by means of an intercalating (i.e. dsDNA-binding) fluorescent green dye, qMax Green, which is similar to SYBR Green, but exhibits higher fluorescence and less PCR inhibition.

qPCR, also known as Real-Time PCR, allows the user to amplify DNA by means of PCR and monitor the progression of amplification quantitatively, in real time. That is, qPCR allows the user to measure quantities of amplified target DNA, cycle by cycle.

qMax Green One Step RT-qPCR Kit contains the following components:

2X Master Mix (containing Accuris Hot Start Taq Polymerase, dNTPs, qMax Green dye, buffer that includes PCR enhancers and inert blue dye that visually aids the user during pipetting)

20X Reverse Transcriptase (an RNA-dependant DNA polymerase, derived from MMLV, and an RNase Inhibitor)

→Available with low or high levels of ROX (select in dropdown menu). ROX is a fluorescent, passive reference dye that is used to normalize the signal emitted from the reporter dye, qMax Green, during qPCR.

The fluorescent signal from the passive dye remains constant during qPCR, while the signal from the reporter dye changes as cycles progress. Real-Time PCR instruments have varying requirements for ROX levels.

Kit is compatible with standard and fast-cycling PCR protocols.

→Available in 10 (sample size), 100, 500 and 1,000 reactions sizes (select in dropdown menu). Reaction sizes are based on total reaction volumes of 20 uL.

Accuris catalog numbers:

Low ROX: 10 rxns (PR2120-L-S), 100 rxns (PR2120-L-100), 500 rxns (PR2120-L-500), 1,000 rxns (PR2120-L-1000)

High ROX: 10 rxns (PR2120-H-S), 100 rxns (PR2120-H-100), 500 rxns (PR2120-H-500), 1,000 rxns (PR2120-H-1000)
